Item A Novel Approach to Prevent Contamination of Domestic Water Using Bacterophage(2020-10-13T10:46:58Z) S, Pradeep; Nisharitha, Ravi; Varsha, P; Mayura, V; Mohammed, MubassirNon availability and lack of access to clean water is one of the major causes of outbreak of diseases. To address this, we have come up with a natural sanitation product acting as a Bio-Disinfectant. We all know, how pure River Ganges is, and it remains pure due to the presence of bacteriophages. By understanding the natural reason behind the purity of River Ganges, our approach towards sanitation product has the ability to kill major bacteria causing diseases like typhoid, cholera, diarrhoea, thus prevents domestic water from getting contaminated and ensures hygiene. Bacteriophage was isolated from sewage source which is rich in bacteria. Presence of bacteriophage was confirmed by plaque assay in pour plate against Escherichia coli and plaque was streak plated to isolate discrete and purified bacteriophage which was stored in SM buffer that maintains the viability of bacteriophage. Preliminary assays were conducted where overnight grown Escherichia coli was added to water sample in different test tubes along with different concentrations of phage and were incubated at room temperature overnight, after which UV Visible spectrophotometric estimation was done at 600nm, and OD was taken at different intervals of time. At a particular concentration of phage, E.coli load was found to decrease which was taken as the optimised value. Optimized concentration of bacteriophage was lyophilized and used effectively to treat water from getting contaminated which was also tested analytically whether the water is fit for drinking. Bacteriophage DNA was further isolated and was given for sequencing (illumina HiSeq) and the data was analysed. Cryo-em was also done to characterize the bacteriophage and to know about proteins on base plate.
